- Calpain-2 is the catalytic subunit of the calcium-activated neutral protease calpain 2, also known as M-calpain or millimolar-calpain, which requires millimolar calcium concentrations for activity.
- It functions as a calcium-regulated, non-lysosomal thiol-protease that mediates limited proteolysis of substrates involved in cytoskeletal remodeling and signal transduction.
- Known substrates include MYOC, proteolytically cleaved at Arg-226, and CPEB3, whose cleavage following neuronal stimulation relieves translational repression of target mRNAs. Related references: PMID:17650508
- Calpain-2 is ubiquitously expressed in human tissues.
- Subcellular localization includes the cytoplasm and association with the cell membrane.
