Biological Background: Calpain 2 Function and Localization

  • Calpain-2 is the catalytic subunit of the calcium-activated neutral protease calpain 2, also known as M-calpain or millimolar-calpain, which requires millimolar calcium concentrations for activity.
  • It functions as a calcium-regulated, non-lysosomal thiol-protease that mediates limited proteolysis of substrates involved in cytoskeletal remodeling and signal transduction.
  • Known substrates include MYOC, proteolytically cleaved at Arg-226, and CPEB3, whose cleavage following neuronal stimulation relieves translational repression of target mRNAs. Related references: PMID:17650508
  • Calpain-2 is ubiquitously expressed in human tissues.
  • Subcellular localization includes the cytoplasm and association with the cell membrane.
  • The protein undergoes post-translational modifications such as acetylation, phosphorylation, and ubiquitination, and features autocatalytic cleavage and isopeptide bond sites.

Experimental Guidance and Technical Tips

  • For Western blot, use standard protocols and consider validating the antibody in your sample system; the predicted molecular weight is ~80 kDa.
  • For immunohistochemistry (paraffin), optimize antigen retrieval conditions and validate in human, mouse, and rat tissues as cross-reactivity is indicated.
  • For ELISA, titrate the antibody to determine the optimal working concentration for your assay format.
  • The immunogen corresponds to the C-terminal region (aa 520-700) of human Calpain 2, a region conserved across species that may account for cross-reactivity with mouse and rat.
