Biological Background: Calpain-2 Catalytic Subunit Function and Localization
- Calpain-2 catalytic subunit (also known as CANP2, mCANP, CANPL2, CANPml) is a calcium-regulated non-lysosomal thiol protease encoded by the CAPN2 gene.
- It catalyzes limited proteolysis of substrates involved in cytoskeletal remodeling and signal transduction.
- Calpain-2 proteolytically cleaves MYOC (myocilin) at Arg-226 (PubMed:17650508). Related references: PMID:17650508
- Following neuronal stimulation, it cleaves CPEB3, abolishing CPEB3 translational repressor activity (By similarity).
- The protein localizes to the cytoplasm and can associate with the cell membrane.
- It is ubiquitously expressed across tissues.
- Key post-translational modifications include acetylation, phosphorylation, and autocatalytic cleavage; the protein also contains calcium-binding domains and isopeptide bonds.
- Calpain-2 is a member of the calpain family and is also designated as M-calpain (millimolar-calpain) due to its calcium requirement.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the C-terminal region (aa 600-700) of human calpain-2. Consider this epitope location when designing validation experiments.
- For Western blotting, a band around 80 kDa corresponds to the full-length catalytic subunit. Validate in relevant human, mouse, or rat sample lysates.
- In ELISA applications, titrate the antibody to optimize signal-to-noise ratios and confirm binding to the immunizing peptide or recombinant protein.
- Given the calcium-dependent activity of calpain-2, consider the impact of calcium concentration on protein conformation if using native protein detection.
- Cross-reactivity with mouse and rat has been reported; validate in your specific sample system.
