Biological Background: Calpain-1 Function and Localization
- Calpain-1, encoded by the CAPN1 gene, is also known as calcium-activated neutral proteinase 1 (CANP 1), mu-calpain, or muCANP, and serves as the catalytic subunit of the micromolar-calpain protease.
- It functions as a calcium-regulated non-lysosomal thiol-protease, mediating limited proteolysis of substrates involved in cytoskeletal remodeling and signal transduction. Related references: PMID:19617626, PMID:21531719, PMID:2400579
- Calpain-1 specifically cleaves CTBP1 at asparagine-375, glycine-387, and histidine-409, and also cleaves and activates caspase-7 (CASP7). Related references: PMID:23707407, PMID:19617626
- Subcellularly, calpain-1 localizes to the cytoplasm and cell membrane.
- It is ubiquitously expressed across human tissues.
- Key features include autocatalytic cleavage, calcium-binding EF-hand domains, and susceptibility to acetylation and phosphorylation; disease-linked mutations in CAPN1 are associated with hereditary spastic paraplegia.
Experimental Guidance and Technical Tips
- For Western blotting, a band of approximately 82 kDa is expected; consider using appropriate positive and negative controls and validating in your sample system.
- For IHC-P, antigen retrieval methods such as heat-induced epitope retrieval may improve detection; optimize staining conditions for your specific tissue type and antibody dilution.
- For IF/ICC, permeabilization is required to detect intracellular calpain-1; fixation conditions should be carefully optimized.
- The antibody is predicted to recognize human, mouse, and rat calpain-1 based on sequence homology; cross-reactivity should be experimentally validated in the relevant model system.
- ELISA performance can be optimized by checkerboard titration of the antibody and antigen; use appropriate blocking and washing steps.
