Biological Background: Calmodulin 1/2/3 Function and Localization

  • Calmodulin is a ubiquitous calcium-binding protein that mediates calcium signal transduction by regulating numerous enzymes, ion channels, and aquaporins. Calcium-binding is required for its activation. Related references: PMID:16760425, PMID:23893133, PMID:26969752
  • It stimulates protein kinases such as myosin light-chain kinases and CaMK2, and phosphatases, playing a central role in calcium-dependent cellular signaling.
  • Calmodulin, together with CCP110 and centrin, regulates the centrosome cycle and progression through cytokinesis. Related references: PMID:16760425
  • Mediates calcium-dependent inactivation of the L-type calcium channel CACNA1C. Related references: PMID:26969752
  • Acts as a positive regulator of calcium-activated potassium channel KCNN2. Related references: PMID:27165696
  • Forms a potassium channel complex with KCNQ1 and regulates its electrophysiological activity. Related references: PMID:25441029
  • Functions as a sensor modulating endoplasmic reticulum contacts with other organelles via VMP1:ATP2A2. Related references: PMID:28890335
  • Component of the SIFI complex required to terminate the mitochondrial stress response. Related references: PMID:38297121, PMID:40328314, PMID:40875847
  • Subcellularly localized to the cytoplasm, cytoskeleton (spindle, spindle pole, microtubule organizing center, centrosome), and cell projections (cilium, flagellum).
  • Required for Legionella pneumophila SidJ glutamylase activity and C. violaceum CopC/S. flexneri OspC3 arginine ADP-riboxanase activity.
  • Key post-translational modifications include acetylation, methylation, phosphorylation, and ubiquitination.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide from amino acids 70–149 of human calmodulin 1/2/3; it may recognize the denatured protein in Western blot under standard reducing conditions.
  • Due to high sequence conservation, the antibody detects calmodulin from human, mouse, and rat; validate in the relevant sample system to confirm specificity.
  • For Western blot, an expected band of approximately 16 kDa should be observed; consider using a high-resolution gel system for clear separation of this small protein.
  • For ELISA, optimize coating antigen concentration and buffer conditions; the antibody is supplied as a polyclonal reagent, so lot-to-lot consistency should be verified in quantitative assays.
  • This antibody is intended for research use only; it has not been validated for diagnostic or therapeutic applications.
