Biological Background: Calmodulin Function and Localization

  • Calmodulin-1, encoded by the CALM1 gene, is a highly conserved calcium-binding messenger protein that transduces intracellular calcium signals.
  • It mediates calcium signal transduction by controlling a large number of enzymes, ion channels, aquaporins, and other proteins through calcium-dependent binding. Related references: PMID:16760425 PMID:23893133 PMID:26969752
  • The calcium-calmodulin complex stimulates various protein kinases, including myosin light-chain kinases and CaMK2, as well as phosphatases. Related references: PMID:16760425 PMID:23893133 PMID:26969752
  • In conjunction with CCP110 and centrin, calmodulin participates in the genetic pathway regulating the centrosome cycle and cytokinesis. Related references: PMID:16760425
  • Calmodulin regulates voltage-dependent L-type calcium channels (mediating calcium-dependent inactivation of CACNA1C) and positively modulates calcium-activated potassium channel KCNN2 activity. Related references: PMID:31454269 PMID:26969752 PMID:27165696
  • It forms a potassium channel complex with KCNQ1, regulating its electrophysiological activity via calcium binding. Related references: PMID:25441029
  • Acts as a sensor to modulate endoplasmic reticulum contacts with other organelles mediated by the VMP1:ATP2A2 complex. Related references: PMID:28890335
  • Subcellularly, calmodulin localizes to the cytoplasm, cytoskeleton (spindle, spindle pole, microtubule organizing center, centrosome), and cell projections (cilium, flagellum).
  • It is a component of the SIFI complex required to resolve mitochondrial stress responses and is also implicated in microbial infection processes (required for legionella SidJ glutamylase activity and bacterial ADP-riboxanase activity). Related references: PMID:38297121 PMID:40328314 PMID:40875847

Experimental Guidance and Technical Tips

  • The immunogen was a synthetic peptide spanning amino acids 70-149 of human Calmodulin (NP_008819.1). This region is highly conserved across Calmodulin isoforms and species, which may explain the reported cross-reactivity with mouse and rat.
  • For Western blot, an expected band around 16 kDa should be observed in positive samples. Consider using calmodulin-enriched tissues or cell lines as positive controls.
  • In immunofluorescence (IF-P), calmodulin may display a diffuse cytoplasmic staining with enrichment at the centrosome, spindle, or ciliary structures. Co-stain with appropriate organelle markers to validate localization.
  • For ELISA, a direct or capture format can be explored, but optimal dilution should be determined empirically using the provided peptide or purified protein.
  • Antibody performance in untested applications or species should be verified independently; consider including relevant positive and negative controls.
