Biological Background: Calmodulin Function and Localization
- Calmodulin-1, encoded by the CALM1 gene, is a highly conserved calcium-binding messenger protein that transduces intracellular calcium signals.

- The immunogen was a synthetic peptide spanning amino acids 70-149 of human Calmodulin (NP_008819.1). This region is highly conserved across Calmodulin isoforms and species, which may explain the reported cross-reactivity with mouse and rat.
- For Western blot, an expected band around 16 kDa should be observed in positive samples. Consider using calmodulin-enriched tissues or cell lines as positive controls.
- In immunofluorescence (IF-P), calmodulin may display a diffuse cytoplasmic staining with enrichment at the centrosome, spindle, or ciliary structures. Co-stain with appropriate organelle markers to validate localization.
- For ELISA, a direct or capture format can be explored, but optimal dilution should be determined empirically using the provided peptide or purified protein.
- Antibody performance in untested applications or species should be verified independently; consider including relevant positive and negative controls.
