Biological Background: Calmodulin 1/2/3 Function and Localization
- Calmodulin-1 (encoded by CALM1), along with the identical calmodulin-2 and calmodulin-3 (encoded by CALM2 and CALM3), is a ubiquitous calcium-binding protein that mediates signal transduction by regulating numerous enzymes, ion channels, and aquaporins in a calcium-dependent manner. Related references: PMID:16760425, PMID:23893133, PMID:26969752
- It is involved in the regulation of the centrosome cycle and cytokinesis together with CCP110 and centrin. Related references: PMID:16760425
- Calmodulin acts as a regulator of voltage-dependent L-type calcium channels and mediates calcium-dependent inactivation of CACNA1C. Related references: PMID:31454269, PMID:26969752
- It positively regulates the calcium-activated potassium channel KCNN2 and forms a complex with KCNQ1 to modulate its electrophysiological activity. Related references: PMID:27165696, PMID:25441029
- Calmodulin functions as a sensor for endoplasmic reticulum interactions with other organelles via the VMP1:ATP2A2 complex. Related references: PMID:28890335
- It is a component of the SIFI complex, which is required to silence the integrated stress response after mitochondrial stress resolution. Related references: PMID:38297121, PMID:40328314, PMID:40875847
- Subcellularly, calmodulin localizes to the cytoplasm, cytoskeleton (spindle, spindle pole, centrosome), and cell projections (cilium, flagellum).
- Post-translational modifications include acetylation, methylation, phosphorylation, isopeptide bond, and ubiquitin conjugation, and it is associated with disease variants in long QT syndrome.
Experimental Guidance and Technical Tips
- This polyclonal antibody is raised against a synthetic peptide spanning amino acids 70-149 of human Calmodulin 1/2/3, a region conserved across the three isoforms; consider validating its specificity in your target species (human, mouse, rat) via Western blot or immunoprecipitation if required.
- Recommended applications include IHC-P, IF/ICC, and ELISA; optimal antibody dilutions should be determined empirically, using positive control tissues or cells known to express calmodulin.
- Given the high conservation of calmodulin, cross-reactivity with other species may occur; if using in non-listed species, perform a homology check and validate with appropriate controls.
- For IHC-P and IF/ICC, antigen retrieval methods and fixation conditions may influence staining; we recommend testing different protocols to achieve the best signal-to-noise ratio.
