Biological Background: CALCOCO1 Function and Localization
- CALCOCO1 (calcium-binding and coiled-coil domain-containing protein 1), also known as calphoglin or coiled-coil coactivator protein, is a multi-functional coactivator that interacts with aryl hydrocarbon and nuclear receptors through its N-terminal domain to regulate transcription.
- It serves as a secondary coactivator in the Wnt/CTNNB1 pathway by binding to CTNNB1 and enhancing LEF1-mediated transcriptional activation, with differential use of activation regions.
- Together with CCAR1, CALCOCO1 enhances GATA1- and MED1-driven transcription from the gamma-globin promoter during erythroid differentiation of K562 cells. Related references: PMID:24245781
- CALCOCO1 also forms part of the calphoglin complex, which links cellular phosphate and glucose metabolism with protein synthesis, degradation, calcium signaling, and cell growth by enhancing inorganic pyrophosphatase activity.
- Subcellular localization studies indicate CALCOCO1 is found in both the cytoplasm and nucleus, consistent with its roles in signaling and transcriptional regulation.
- The protein contains zinc-finger motifs and is phosphorylated, suggesting regulatory modifications; alternative splicing generates multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen used for this polyclonal antibody corresponds to amino acids 462-691 of human CALCOCO1 (NP_065949.1). The resulting antiserum is expected to recognize the full-length protein and may detect isoforms containing this region.
- For Western blotting, consider using whole-cell lysates from human or mouse cell lines known to express CALCOCO1 (e.g., K562 for the gamma-globin regulatory function). A band near 77 kDa is anticipated.
- ELISA applications may benefit from optimization of coating antigen concentration and antibody dilution to achieve desired sensitivity.
- Cross-reactivity with other species or isoforms should be validated in the relevant sample system, as the antibody has only been tested on human and mouse samples.
