- CADM3 (Cell adhesion molecule 3), also known as NECL1, SynCAM3, IGSF4B, or TSLL1, is a member of the immunoglobulin superfamily and nectin-like family of adhesion molecules. It is expressed primarily in the brain, with isoform 1 in adult and fetal brain and isoform 2 highly in adult brain cerebellum and weakly in placenta.
- It mediates calcium-independent homophilic cell-cell adhesion and heterophilic interactions with nectin-1, nectin-3, and IGSF4/CADM1, contributing to synaptic organization and cell junction stability.
- CADM3 localizes to the cell membrane and cell junctions, where it may interact with EPB41L1 (erythrocyte membrane protein band 4.1-like 1) to regulate junctional structure and function (By similarity).
- Post-translational modifications include glycosylation and phosphorylation, and the protein contains transmembrane and immunoglobulin-like domains essential for adhesion.
- Alternative splicing generates multiple isoforms, and mutations in CADM3 are associated with Charcot-Marie-Tooth disease type 2FF (CMT2FF), highlighting its role in peripheral nerve integrity and neurodegeneration.
