  • CAD is a multifunctional protein that carries out the first three enzymatic activities of the de novo pyrimidine biosynthesis pathway: carbamoylphosphate synthetase (CPSase), aspartate transcarbamylase (ATCase), and dihydroorotase (DHOase).
  • The CPSase function occurs in two steps: a glutamine-dependent amidotransferase (GATase) domain generates ammonia, which is then used by the ammonium-dependent carbamoyl phosphate synthetase domain to produce carbamoyl phosphate from ATP and hydrogencarbonate.
  • ATCase catalyzes the formation of carbamoyl-L-aspartate from L-aspartate and carbamoyl phosphate.
  • DHOase catalyzes the cyclization of carbamoyl aspartate to dihydroorotate, completing the initial steps of UMP synthesis.
  • CAD localizes to both the cytoplasm and the nucleus.
  • It is subject to acetylation and phosphorylation, and functions as a zinc- and metal-dependent allosteric enzyme.
  • Mutations in CAD are associated with congenital disorder of glycosylation type 1Z (CDG1Z) and developmental and epileptic encephalopathy 50 (DEE50/EIEE50).
