Biological Background: CACNB1 Function and Localization
- CACNB1 encodes the voltage-dependent L-type calcium channel subunit beta-1, a regulatory subunit that modulates channel gating, trafficking, and surface expression. Related references: PMID:1309651, PMID:15615847, PMID:8107964
- It is required for functional expression and activity regulation of L-type channels containing pore-forming subunits CACNA1C (PubMed:15615847), CACNA1D (PubMed:1309651), and CACNA1B (PubMed:8107964); regulation of CACNA1A-containing channels is inferred by similarity.
- Localizes to the cell membrane and sarcolemma, ensuring proper channel clustering at the plasma membrane.
- Isoform 1 and isoform 3 are expressed in brain, heart, spleen, central nervous system, and neuroblastoma cells; isoform 2 is predominantly found in skeletal muscle. Detectable at the protein level in heart ventricle. Related references: PMID:15615847
- Post-translational modifications include phosphorylation (classified as a phosphoprotein) and alternative splicing, generating multiple functional isoforms with distinct tissue distributions.
Experimental Guidance and Technical Tips
- For western blot analysis, consider using protein lysates from human heart or brain tissues, as CACNB1 is robustly expressed in these organs. Mouse and rat samples are also suitable for cross-reactivity validation.
- The immunogen spans residues 429–598, located in the C-terminal region; due to alternative splicing, the antibody may detect multiple isoforms, potentially leading to additional bands.
- For ELISA applications, validate binding using the recombinant immunogen or full-length protein to confirm specificity and optimize coating conditions.
