Biological Background: CACNA2D3 Function and Localization
- CACNA2D3, also known as voltage-gated calcium channel subunit alpha-2/delta-3, is the alpha-2/delta-3 subunit of voltage-dependent calcium channels.
- It regulates calcium current density and activation/inactivation kinetics of the calcium channel, acting as a regulatory subunit for P/Q-type (CACNA1A), N-type (CACNA1B), and L-type (CACNA1C or CACNA1D) channels, but not T-type (CACNA1G).
- At the protein level, CACNA2D3 is only detected in brain, with no expression in lung, testis, aorta, spleen, jejunum, ventricular muscle, and kidney. Related references: PMID:11687876 PMID:11245980
- The protein is a membrane protein with a signal peptide, transmembrane helix, and multiple disulfide bonds, glycosylation, and phosphorylation sites.
- Keywords from UniProt include alternative splicing, calcium channel, calcium transport, glycoprotein, ion channel, ion transport, metal-binding, phosphoprotein, and voltage-gated channel, indicating diverse functions and modifications.
- The CACNA2D3 gene is a member of the CACNA2D family, which comprises four genes encoding auxiliary subunits of voltage-gated calcium channels.
Experimental Guidance and Technical Tips
- The immunogen comprises amino acids 900-1040 of human CACNA2D3. This region may be present in full-length and certain splice variants; consider validating antibody specificity with sequence alignment and appropriate positive controls.
- For Western blotting, the predicted molecular weight is ~123 kDa. Use a suitable gel percentage (e.g., 6-8% SDS-PAGE) and optimize transfer conditions for high-molecular-weight proteins.
- Since CACNA2D3 is brain-specific, use brain tissue lysates or relevant cell lines to confirm antibody performance in WB.
- For ELISA applications, titrate the antibody to determine the optimal working concentration for your assay.
