Biological Background: CACNA2D2 Function and Localization
- Encodes the alpha-2/delta-2 subunit of voltage-dependent calcium channels, also annotated as voltage-gated calcium channel subunit alpha-2/delta-2.
- Regulates calcium current density and activation/inactivation kinetics of P/Q-type (CACNA1A), N-type (CACNA1B), L-type (CACNA1C or CACNA1D), and possibly T-type (CACNA1G) calcium channels. Related references: PMID:15111129 PMID:23339110
- Overexpression of CACNA2D2 induces apoptosis.
- Predominantly expressed in cerebellar cortex; also detected at high levels in heart, lung, testis, pancreas, and skeletal muscle, with lower expression in kidney, liver, placenta, and brain. Notably present in lung tumor cell lines but absent in normal lung at the protein level.
- Localized to the membrane; features a signal peptide, N-glycosylation, and disulfide bonds.
- Subject to alternative splicing; associated with disease variants in epilepsy.

- The immunogen comprises amino acids 20-200 of human CACNA2D2, corresponding to an extracellular N-terminal region; consider this epitope location when designing detection or functional assays.
- For Western blot (WB), the predicted molecular weight is ~130 kDa. Use a low-percentage SDS-PAGE gel (e.g., 6-8%) and optimize transfer conditions for high-molecular-weight proteins.
- Cross-reactivity with mouse and rat extends utility to rodent models; validate with appropriate positive and negative controls in your experimental system.
- Expression of CACNA2D2 varies across tissues; perform tissue-specific validation, particularly if working with cerebellar, cardiac, or lung samples.
