- CACNA1S, also known as Cav1.1, is the pore-forming alpha-1S subunit of the voltage-gated L-type calcium channel in skeletal muscle.
- It mediates L-type calcium currents and is critical for excitation-contraction coupling by interacting with RYR1 to trigger Ca(2+) release from the sarcoplasmic reticulum.
- This channel belongs to the high-voltage activated (HVA) group of calcium channels.
- CACNA1S is specifically localized to the sarcolemma and T-tubule membranes of skeletal muscle cells.
- Expression is restricted to skeletal muscle tissue.
- The protein contains multiple transmembrane domains, is glycosylated, and is regulated by calmodulin binding and phosphorylation.
- Mutations in CACNA1S are associated with diseases such as hypokalemic periodic paralysis and malignant hyperthermia susceptibility.
