Biological Background: CABP1 Function and Localization
- CABP1 (Calcium-binding protein 1), also known as calbrain or caldendrin, is a neuronal calcium sensor protein involved in sensory transduction, synapse function, and vision. The gene undergoes alternative splicing, producing multiple isoforms including the brain-specific calbrain.
- Modulates calcium-dependent activity of inositol 1,4,5-triphosphate receptors (ITPRs). Related references: PMID:14570872
- Inhibits agonist-induced intracellular calcium signaling. Related references: PMID:15980432
- Regulates voltage-gated calcium channels: enhances inactivation of P/Q-type channels and causes calcium-dependent facilitation while inhibiting inactivation of L-type channels by binding to CACNA1C. Related references: PMID:11865310, PMID:15140941
- Suppresses the calcium-dependent inactivation of CACNA1D.
- Inhibits TRPC5 channels. Related references: PMID:15895247
- Prevents NMDA receptor-induced cellular degeneration and is required for the normal transfer of light signals through the retina.
- Exhibits a broad subcellular distribution including cytoplasm, cytoskeleton, cell membrane, Golgi, and postsynaptic density; predominantly expressed in brain (hippocampus, habenular area, cerebellum) and retina, with calbrain isoform restricted to brain.
Experimental Guidance and Technical Tips
- The immunogen was a synthetic peptide spanning amino acids 1–100 of human CABP1 (accession NP_001028849.1). This N-terminal region is expected to be well-exposed in the native protein.
- For Western blotting, the predicted molecular weight is ~40 kDa. Due to alternative splicing and post-translational modifications, additional bands may be observed in some sample types.
- In immunohistochemistry (paraffin), CABP1 is known to be highly expressed in brain and retina; ensure appropriate positive control tissues such as hippocampus or cerebellum are included.
- For ELISA applications, consider using this polyclonal antibody as either capture or detection reagent, and validate performance with recombinant CABP1 protein.
- The antibody has been reported to cross-react with mouse and rat CABP1 based on sequence homology; experimental validation in the relevant species is recommended.
