Biological Background: C9 Function and Localization

  • Functions as the pore-forming subunit of the membrane attack complex (MAC), which inserts into target cell membranes to induce lysis. Related references: PMID:22832194, PMID:26841837, PMID:26841934
  • MAC assembly is triggered by proteolytic cleavage of C5 into C5b, which can be initiated by the classical, alternative, lectin or GZMK complement pathways. Related references: PMID:39914456, PMID:39814882, PMID:9212048
  • The complement cascade enhances phagocytosis, pathogen breakdown, and signaling that strengthens the adaptive immune system. Related references: PMID:9212048, PMID:9634479
  • During MAC formation, C9 first binds the C5b8 complex and then polymerizes to form the final transmembrane pore. Related references: PMID:26841934, PMID:30111885, PMID:30552328
  • C9 is secreted and inserts into target cell membranes; it is predominantly detected in plasma.
  • The protein undergoes post-translational modifications including glycosylation and phosphorylation, and contains EGF-like domains and disulfide bonds that contribute to its structure.
  • Mutations in C9 are associated with age-related macular degeneration (ARMD15).

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 22–265 of human C9; cross-reactivity with mouse C9 has been reported but should be validated experimentally if precise reactivity is critical.
  • For Western blotting, the predicted molecular weight is 63 kDa; however, post-translational modifications may cause shifts, so optimization of blocking and detection conditions is recommended.
  • In ELISA applications, include appropriate antigen and negative controls to confirm specificity, and consider checkerboard titrations for optimal antibody dilution.
  • Store lyophilized or reconstituted antibody at −20°C or below, and aliquot to avoid repeated freeze-thaw cycles.
