Biological Background: G6b Function and Localization
- Encoded by the MPIG6B gene, G6b (also known as C6orf25, G6b-B, NG31, THAMY) is a megakaryocyte and platelet inhibitory receptor that belongs to the immunoglobulin superfamily.
- Acts as a critical regulator of hematopoietic lineage differentiation, megakaryocyte function, and platelet production by inhibiting platelet aggregation and activation in response to agonists such as ADP and collagen-related peptide (PubMed:12665801). Related references: PMID:12665801
- Contains two immunoreceptor tyrosine-based inhibitory motifs (ITIMs) in its cytoplasmic tail, through which it inhibits CLEC1B and GP6:FcRgamma signaling pathways to regulate platelet function (PubMed:17311996, PubMed:27743390). Related references: PMID:17311996, PMID:27743390
- Isoform B is the only isoform containing both a transmembrane region and two ITIMs; isoform A may act as an activating counterpart.
- Subcellularly localizes to the endoplasmic reticulum, Golgi apparatus, and cell membrane.
- Expressed primarily in platelets and restricted hematopoietic cell lines (K-562, MOLT-4, Jurkat), but not detected in monocyte, B-cell, fibroblast, or natural killer cell lines.
- Post-translationally modified by phosphorylation (phosphoprotein) and contains a heparin-binding domain.
- Keywords highlight its roles as a glycoprotein, heparin-binding protein, and transmembrane signaling receptor.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the extracellular domain (amino acids 18–142) of human C6orf25, which is conserved across isoforms and species; this may enable detection of both isoform A and B in human, mouse, and rat samples.
- For western blotting, a predominant band around 26 kDa is expected in human platelet or megakaryocyte lysates; validate the exact migration pattern in your experimental system.
- Cross-reactivity with mouse and rat allows for potential use in rodent models; however, verify specificity in the relevant tissue or cell line.
- In ELISA applications, this polyclonal antibody can be used as a capture or detection reagent; consider optimizing coating concentration and pairing with a specific monoclonal antibody for sandwich ELISA formats.
