Biological Background: C5AR2 Function and Localization
- C5AR2, also known as C5L2, GPF77, and GPR77, is a G protein-coupled receptor (GPCR) that binds anaphylatoxins C5a and their dearginated forms (ASP/C3adesArg, C4adesArg, C5adesArg).
- Upon C5a binding, C5AR2 stimulates chemotaxis, granule enzyme release, intracellular calcium release, and superoxide anion production. Related references: PMID:11773063
- C5AR2 signals via G(i)/G(o) G alpha proteins (GNAI1 or GNAO1), leading to inhibition of adenylate cyclase. Related references: PMID:12540846
- The receptor also recognizes dearginated peptides ASP/C3adesArg, C4adesArg, and C5adesArg, with ligand binding triggering a conformational change modulating downstream effectors. Related references: PMID:12540846 PMID:15833747 PMID:19615750
- Subcellularly, C5AR2 is a multi-pass membrane protein that localizes to the cell membrane.
- Tissue expression includes frontal cortex, hippocampus, hypothalamus, pons, and liver.
- The protein features disulfide bonds, glycosylation, and phosphorylation sites as annotated in UniProt.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 258-337) of human C5AR2, suggesting specific recognition of the receptor's intracellular domain.
- For Western blot (WB), the predicted molecular weight is 36 kDa; however, due to post-translational modifications such as glycosylation and phosphorylation, the observed band may migrate at a higher apparent molecular weight. Validate using appropriate positive controls (e.g., C5AR2-expressing cell lines).
- In ELISA applications, the antibody can be employed for both direct and indirect detection of the C5AR2 protein or its fragments. Optimize coating and detection conditions for best signal-to-noise ratio.
- The antibody shows cross-reactivity with both human and mouse C5AR2, making it suitable for translational studies. Always verify specificity in your experimental system.
- Consider using this polyclonal antibody in conjunction with C5AR2 agonists (e.g., C5a) or antagonists to study receptor activation and downstream signaling.
