Biological Background: Complement C4-B Function and Localization
- Complement C4-B is a central component of the classical and lectin complement pathways, functioning as a precursor that is proteolytically activated to drive opsonization, pathogen clearance, and adaptive immune signaling.
- Following cleavage by C1S, MASP2, or GZMK, the C4b fragment covalently attaches to pathogen surfaces and recruits C2 to form the C3 and C5 convertases. The C4b-B isotype preferentially catalyzes transacylation of its thioester carbonyl group to form ester bonds with carbohydrate antigens, while the C4b-A isotype forms amide bonds with protein antigens. - During activation, the anaphylatoxin-like fragment C4a is released; its role in local inflammation is controversial, as early reports may have been confounded by contaminating C3a and C5a.
- Complement C4-B is primarily expressed in the liver, with moderate levels in the adrenal cortex, adrenal medulla, thyroid gland, and kidney, and lower levels in the heart, ovary, small intestine, thymus, pancreas, and spleen, suggesting both systemic and local immune functions.
- Subcellularly, C4-B is secreted and can also be found at cell surfaces, synapses, and neuronal projections (axons and dendrites), indicating potential roles in synaptic pruning and neuroinflammation.
- The protein undergoes extensive post-translational modifications including glycosylation, sulfation, and phosphorylation, and contains a critical thioester bond essential for covalent attachment to target surfaces.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C4c fragment (residues 1337–1446); the antibody is expected to recognize both full-length C4 and the cleaved C4c fragment. Validate specificity in your sample system.
- For western blot, the predicted molecular weight of full-length C4 is ~192 kDa; under reducing conditions, multiple bands corresponding to the alpha, beta, and gamma chains may be observed. Consider optimizing protein loading and detection conditions.
- In immunofluorescence applications (IF/ICC, IF-P), permeabilization and fixation protocols should be tailored to detect secreted and membrane-associated C4.
- The reported cross-reactivity with mouse and rat enables use in rodent models; confirm reactivity in your tissue or cell type of interest.
