Biological Background: Complement C3 Function and Localization

• Complement C3 is a central precursor of non-enzymatic components in the classical, alternative, lectin, and GZMK complement pathways, leading to opsonization, phagocytosis, and immune signaling.
• Upon cleavage by C3 convertase, C3b is generated and covalently attaches to pathogen surfaces via its reactive thioester bond, acting as an opsonin and forming part of the C5 convertase by recruiting complement C2b or CFB.
• The anaphylatoxin C3a, released through cleavage, binds to the C3AR1 receptor to stimulate mast cell and basophil degranulation, promoting inflammation and acting as a chemoattractant for macrophages and neutrophils.
