Biological Background: Complement C3 Function and Localization
- Complement C3 is a central precursor of non-enzymatic components in the classical, alternative, lectin, and GZMK complement pathways, leading to opsonization, phagocytosis, and immune signaling.
- Upon cleavage by C3 convertase, C3b is generated and covalently attaches to pathogen surfaces via its reactive thioester bond, acting as an opsonin and forming part of the C5 convertase by recruiting complement C2b or CFB.
- The anaphylatoxin C3a, released through cleavage, binds to the C3AR1 receptor to stimulate mast cell and basophil degranulation, promoting inflammation and acting as a chemoattractant for macrophages and neutrophils.
- An adipogenic fragment of C3 (ASP/C3adesArg) stimulates triglyceride synthesis and glucose transport in adipocytes via C5AR2, regulating fat storage without pro-inflammatory activity.
- In chronic inflammation, C3-derived peptides act as chemoattractants for neutrophils, facilitating migration to inflamed tissues.

- The immunogen corresponds to a peptide within amino acids 1159-1286 of mouse C3, a region within the C3d domain; this design may favor recognition of the native protein and its cleaved fragments.
- For Western blotting, consider using reducing conditions and appropriate molecular weight standards to detect the 186 kDa precursor or processed chains. Validate specificity using relevant mouse or rat serum or tissue lysates that express C3.
- In ELISA applications, the polyclonal nature of this antibody may enhance detection sensitivity; however, cross-reactivity with other complement components should be assessed experimentally.
