Biological Background: C1S Function and Localization
- Complement C1s subcomponent (also known as C1 esterase) is a serine protease that forms part of the C1 complex, which initiates the classical complement pathway upon binding to antigen-antibody complexes on pathogen surfaces. Related references: PMID:11445589 PMID:16169853 PMID:417728 PMID:34155115
- C1S is activated through cleavage by C1R, generating heavy and light chains. Related references: PMID:11445589 PMID:6271784
- The catalytic light chain of C1S subsequently cleaves and activates complement components C2 and C4, propagating the classical pathway. Related references: PMID:16169853 PMID:467643 PMID:6282646
- C1S also cleaves other substrates, including C1 inhibitor (SERPING1; in vitro evidence) and IGFBP5, thereby modulating IGF1 activity. Related references: PMID:16169853 PMID:18930415
- C1S is secreted and can be found at the cell surface, consistent with its role in the extracellular complement cascade.
- The protein is characterized by several structural features, including calcium-binding sites, EGF-like domains, and disulfide bonds, and it undergoes hydroxylation and glycosylation. Mutations in C1S are associated with Ehlers-Danlos syndrome.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 479–688 of human C1S, encompassing the light chain region; the antibody may recognize both the full-length proenzyme and the activated light chain. For Western blot, consider running samples under reducing and non-reducing conditions to distinguish cleaved and uncleaved forms.
- This polyclonal antibody is suitable for WB and ELISA; for ELISA, optimal detection may require validation in serum or plasma samples where secreted C1S is present.
- Cross-reactivity with rat is reported; validate in the relevant species and sample matrix to ensure specificity.
- No validated immunohistochemistry or other applications are provided; verify before use.
