Biological Background: C1QA Function and Localization
- C1QA encodes the A chain of complement C1q, a collagen-like and globular domain-containing protein that assembles with C1QB and C1QC to form the hexameric C1q subcomponent of the C1 complex, the first component of the classical complement pathway. Related references: PMID:12847249, PMID:19006321, PMID:24626930
- The C1q subcomponent specifically binds to the Fc regions of IgG and IgM antibodies when they are complexed with antigens, triggering activation of the proenzymes C1R and C1S and initiating the proteolytic cascade that eliminates pathogens and enhances adaptive immunity. Related references: PMID:29449492, PMID:19706439, PMID:24626930
- Beyond immune complex recognition, C1q also binds phosphatidylserine exposed on apoptotic cells, facilitating clearance of cell debris and possibly promoting complement activation in a non-immunoglobulin-dependent manner. Related references: PMID:18250442
- C1QA is a secreted protein that can also be found on the cell surface, consistent with its role in extracellular immune surveillance.
- The protein contains collagen-like repeats, disulfide bonds, and is subject to hydroxylation and glycosylation, which are critical for its structural stability and interaction with immune complexes.
- C1QA has a predicted molecular weight of approximately 26 kDa (UniProt: 26017 Da), though it may run at higher observed weights due to post-translational modifications; the immunogen described here is a recombinant fragment corresponding to amino acids 23-245 of the human C1QA precursor.
Experimental Guidance and Technical Tips
- The recombinant immunogen spanning amino acids 23–245 of human C1QA likely includes the collagen-like and globular C1q domains; consider whether target epitopes may be masked in native complexes if using native sample preparations.
- This antibody shows broad species cross-reactivity (human, mouse, rat); validate in the relevant sample system and consider potential interference from heterophilic antibodies in species-mismatched assays.
- WB and ELISA applications are supported; for WB, the predicted band size is ~26 kDa, but higher molecular weight species may be detected due to multimerization or glycosylation; using reducing and denaturing conditions may resolve distinct bands.
- In ELISA formats, ensure proper coating of the recombinant antigen or C1q complex; note that the antibody may recognize both free and complexed C1QA.
