- C1GALT1C1 (also known as COSMC, C1Gal-T2, C38H2-L1) is a ubiquitously expressed molecular chaperone essential for the biosynthesis of O-glycans, specifically the T antigen (Gal-beta1-3GalNAc-alpha1-Ser/Thr).
- It acts by assisting the folding and stability of core 1 beta-3-galactosyltransferase (C1GALT1), enabling the generation of core 1 O-glycan structures that serve as precursors for many extended O-glycans in glycoproteins.
- The protein is a membrane-associated chaperone with a signal-anchor and a predicted transmembrane helix, consistent with its localization at the endoplasmic reticulum/Golgi interface where O-glycosylation occurs.
- Tissue profiling shows abundant expression in salivary gland, stomach, small intestine, kidney, and testis, with moderate levels in brain, cerebellum, thymus, spleen, lung, pancreas, ovary, and uterus.
- Mutations in C1GALT1C1 are linked to disease variants, including atypical hemolytic uremic syndrome, highlighting its importance in glycoprotein quality control.
