Biological Background: c-Maf Function and Localization
- c-Maf (MAF) is a basic leucine zipper (bZIP) transcription factor belonging to the Maf family, acting as both a transcriptional activator and repressor. It is alternatively spliced to generate multiple isoforms.
- It plays a critical role in embryonic lens fiber cell development by recruiting the transcriptional coactivators CREBBP and EP300 to crystallin gene promoters, thereby up-regulating crystallin expression.
- In T helper 2 (Th2) cells, c-Maf activates IL4 expression and modulates T-cell susceptibility to apoptosis by interacting with MYB and decreasing BCL2 expression.
- c-Maf, together with PAX6, transactivates the glucagon gene promoter; it also regulates CD13 expression in endothelial cells and during myelopoiesis, either activating or repressing transcription depending on the cell context.
- The protein localizes to the nucleus and is predominantly expressed in endothelial cells.
- It can function either as an oncogene or a tumor suppressor depending on the cellular environment, and it represses antioxidant response element (ARE)-mediated transcription when overexpressed.
- UniProt keywords highlight its involvement in disease variants such as cataract, deafness, intellectual disability, and dwarfism, as well as its role as a proto-oncogene and tumor suppressor.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fragment corresponding to the C-terminal region (amino acids 314-403) of human c-Maf, which may allow detection of multiple isoforms; confirm specificity in your sample system.
- For western blotting, the predicted molecular weight of full-length c-Maf is ~38 kDa; use appropriate positive controls such as endothelial cell lysates or lens tissue extracts.
- The antibody cross-reacts with mouse and rat c-Maf, making it suitable for studies involving rodent models; validate performance in these species using appropriate controls.
- For ELISA, consider using recombinant c-Maf protein or cell lysates as coating antigens, and optimize concentrations based on your assay conditions.
