Biological Background: c-Jun Function and Localization

  • c-Jun (AP-1, p39) is a transcription factor and a core component of the AP-1 complex, belonging to the basic leucine zipper (bZIP) family.
  • It recognizes and binds the AP-1 consensus motif 5'-TGA[GC]TCA-3' to regulate gene expression (PubMed:10995748, PubMed:22083952). Related references: PMID:10995748 PMID:22083952
  • Heterodimerizes with FOS family proteins to form an active AP-1 transcription complex, enhancing DNA binding and transcriptional activity (By similarity).
  • Plays a role in activation-induced cell death of T cells by binding to the FASLG/CD95L promoter and inducing its transcription (PubMed:12618758). Related references: PMID:12618758
  • Promotes NR5A1 activity when phosphorylated by HIPK3, leading to increased steroidogenic gene expression upon cAMP signaling (PubMed:17210646). Related references: PMID:17210646
  • Involved in activated KRAS-mediated transcriptional activation of USP28 in colorectal cancer cells (PubMed:24623306). Related references: PMID:24623306
  • Localizes predominantly to the nucleus and is expressed in the developing and adult prostate as well as prostate cancer cells.
  • Post-translationally modified by phosphorylation and acetylation, which regulate its transcriptional activity.

Experimental Guidance and Technical Tips

  • The immunogen spans the N-terminal region (aa 1–70) of c-Jun; ensure the epitope is accessible in your assay format.
  • For Western blot, the predicted molecular weight is 36 kDa; consider that post-translational modifications may cause band shifts.
  • This antibody shows cross-reactivity with mouse and rat, but validation in your specific experimental system is recommended.
  • ELISA applications may require optimization of coating conditions and detection reagents.
