Biological Background: bZIP46 Function and Localization
- bZIP46, also known as OsAREB8, OsABF2, OsABF3, and ABI5-LIKE 1, is a basic leucine zipper (bZIP) transcription factor in rice (Oryza sativa).
- It functions as a key component in abscisic acid (ABA) signaling; its transcription factor activity is fully activated by ABA treatment.
Related references:
PMID:19947981, PMID:26300907, PMID:27468891 - bZIP46 positively regulates the expression of abiotic stress-responsive genes through an ABA-dependent signaling pathway, thereby enhancing drought stress tolerance.
Related references:
PMID:20576316, PMID:22301130 - The protein is involved in crosstalk between ABA and auxin signaling: it directly binds to ABA-responsive elements (ABREs) in target gene promoters, notably WRKY family genes, and suppresses auxin signaling by targeting ABRE-containing genes related to auxin metabolism or signaling.
Related references:
PMID:21546455 - bZIP46 localizes to the nucleus, consistent with its role as a transcription factor.
- It is widely expressed in various tissues, including roots, shoots, leaves, flag leaves, stems, flowers, and panicles.
Related references:
PMID:20576316, PMID:21546455 - As a member of the AREB/ABF family, bZIP46 contains a conserved bZIP domain required for DNA binding and dimerization; it is classified under stress response and transcription regulation keywords.
Experimental Guidance and Technical Tips
- The immunogen comprises a recombinant protein fragment (amino acids 100–200), which spans a central region of bZIP46. For Western blot (WB), we recommend using whole-cell lysates from Oryza sativa tissues, with detection expected at approximately 35 kDa. Due to potential post-translational modifications, the observed band may shift slightly.
- For ELISA, consider using purified recombinant bZIP46 or bZIP46-derived peptides as coating antigens. Include appropriate negative and positive controls to validate assay specificity.
- As a polyclonal antibody, lot-to-lot variability may occur; we recommend titrating the antibody for each new application and validating performance in the relevant sample system.
- Cross-reactivity is reported for Oryza sativa; testing in other plant species may require sequence homology analysis of the immunogen region.
