Biological Background: BYSL Function and Localization
- Bystin (encoded by BYSL/ENP1 gene) is a 50 kDa protein involved in ribosome biogenesis and cell adhesion.
- Essential for processing of 20S pre-rRNA precursor into mature 40S ribosomal subunits.
- May mediate trophinin-dependent cell adhesion during human embryo implantation.
- Localizes to the cytoplasm, nucleus, and nucleolus, consistent with its role in ribosomal processing.
- Expressed in placenta from week 6 of pregnancy, predominantly in syncytiotrophoblast and endometrial decidual cells; levels decline after week 10.
- Post-translational modifications include phosphorylation and methylation (UniProt keyword annotations).
- Identified in proteomics screens, supporting its role in fundamental cellular processes (Reference proteome).
Experimental Guidance and Technical Tips
- The immunogen corresponds to a central region (aa 120-280) of human BYSL; the antibody may recognize both full-length protein and potential isoforms.
- For western blotting, the predicted band size is ~50 kDa; consider using positive control lysates from HeLa cells or placental tissue.
- In paraffin-embedded immunohistochemistry, antigen retrieval may be required; validate with relevant tissue sections such as first-trimester placenta.
- ELISA performance should be optimized with appropriate coating antigen and detection conditions.
