Biological Background: BVES Function and Localization

  • BVES (Popeye domain-containing protein 1, also known as POPDC1) is a cell adhesion molecule that plays a key role in establishing and maintaining cell integrity. It is involved in the formation and regulation of tight junction (TJ) paracellular permeability in epithelial cells. Related references: PMID:16188940
  • The protein interacts with VAMP3, modulating VAMP3-mediated vesicular transport and the recycling of various receptor molecules. It also regulates cell shape and movement by modulating Rho-family GTPase activity through interaction with ARHGEF25/GEFT.
  • BVES induces primordial adhesive contact and aggregation of epithelial cells in a Ca²⁺-independent manner. It is also implicated in striated muscle regeneration, repair, and the regulation of cell spreading (By similarity).
  • In cardiac muscle, BVES is essential for maintaining cardiac function. It regulates heart rate dynamics, at least in part, through cAMP-binding and by enhancing cell surface expression of the potassium channel KCNK2, thereby increasing current density. Related references: PMID:26642364
  • BVES localizes to the lateral cell membrane, tight junctions, sarcolemma, and caveolae. Its caveolae association is critical for preserving the structural and functional integrity of caveolae and protecting the heart against ischemia injury.

  • The immunogen corresponds to the C-terminal region (aa 241-360) of human BVES. This region may provide good antigenicity; researchers should consider its accessibility in native versus denaturing conditions.
  • For Western blot (WB), a band at approximately 41 kDa is expected. Include appropriate positive controls, such as lysates from human heart or skeletal muscle tissue, or epithelial cell lines known to express BVES.
  • In ELISA applications, optimize coating concentration and block non-specific binding carefully. Recombinant BVES protein or cell lysates with confirmed expression can be used as target antigens.
  • Due to the role of BVES in tight junctions and caveolae, subcellular fractionation or specific lysis conditions may improve detection. Validate the antibody‘s performance in the relevant cellular context.
  • Cross-reactivity has been verified for human; if using in other species, confirm sequence homology and perform preliminary validation.
