Biological Background: BUB1 Function and Localization
- BUB1 is a serine/threonine-protein kinase essential for spindle-assembly checkpoint signaling and proper chromosome alignment during mitosis.
- It is required for the kinetochore recruitment of multiple checkpoint proteins, including CENPF, BUB1B, CENPE, MAD2L1, and PLK1, and influences AUKRB localization.
- BUB1 promotes centromeric localization of TOP2A and defines SGO1 localization, thereby regulating sister chromatid cohesion. Related references: PMID:35044816
- As a substrate of APC/C-Cdh1, the BUB1-BUB3 complex phosphorylates CDC20 to inhibit the anaphase-promoting complex/cyclosome, ensuring mitotic checkpoint activity.
- BUB1 mediates apoptosis in response to chromosome missegregation and acts as a tumor suppressor by preventing spontaneous tumorigenesis.
- Expressed predominantly in tissues with a high mitotic index, such as testis and thymus, and localizes to the nucleus, centromere, and kinetochore.
Experimental Guidance and Technical Tips
- The immunogen covers the N-terminal region (aa 1–300) of human BUB1; therefore, the antibody may recognize both full-length and N-terminal fragments. Consider this when interpreting band patterns.
- For Western blotting, a predicted band size of 122 kDa suggests using a medium-percentage SDS-PAGE gel (e.g., 8%) and optimized transfer conditions for high-molecular-weight proteins.
- The antibody is validated for WB and ELISA, and cross-reacts with human and mouse samples. When using other species or applications, validate in the relevant sample system.
