  • BUB1 is a serine/threonine-protein kinase essential for spindle-assembly checkpoint signaling and proper chromosome alignment during mitosis.
  • It is required for the kinetochore recruitment of multiple checkpoint proteins, including CENPF, BUB1B, CENPE, MAD2L1, and PLK1, and influences AUKRB localization.
  • BUB1 promotes centromeric localization of TOP2A and defines SGO1 localization, thereby regulating sister chromatid cohesion. Related references: PMID:35044816
  • As a substrate of APC/C-Cdh1, the BUB1-BUB3 complex phosphorylates CDC20 to inhibit the anaphase-promoting complex/cyclosome, ensuring mitotic checkpoint activity.
