Biological Background: BTG3 Function and Localization
- Protein BTG3, also known as abundant in neuroepithelium area protein (ANA) or Tob5, is a member of the BTG/Tob family of antiproliferative factors.
- Overexpression of BTG3 impairs serum-induced cell cycle progression from the G0/G1 to S phase, suggesting a role as a negative regulator of cell proliferation.
- BTG3 is ubiquitously expressed, with high levels in the ventricular zone of the developing central nervous system and in adult tissues including ovary, testis, prostate, thymus, and lung.
- The BTG3 gene undergoes alternative splicing, producing multiple transcript variants.
- The protein has been identified in large-scale proteomics studies and is included in the human reference proteome.
- The calculated molecular weight of BTG3 is approximately 29 kDa (29116 Da), consistent with the observed ~29 kDa band.
Experimental Guidance and Technical Tips
- This polyclonal antibody was generated against a recombinant protein spanning amino acids 150–250 of human BTG3; verify reactivity in the relevant sample system prior to use.
- For Western blot, a specific band near 29 kDa is expected; consider using molecular weight markers and, if available, a knockdown or knockout control to confirm specificity.
- In ELISA, titrate the antibody against the immunogen or a known BTG3-containing sample to determine optimal working concentration.
- Cross-reactivity is reported for human BTG3; for other species, sequence homology should be examined and validity assessed experimentally.
