Biological Background: BTC Function and Localization

  • Probetacellulin (BTC) is a member of the epidermal growth factor (EGF) family, synthesized as a transmembrane precursor that is proteolytically cleaved to release the mature growth factor.
  • BTC functions as a high-affinity ligand for EGFR, ERBB4, and other EGF receptor family members, acting as a potent mitogen for retinal pigment epithelial cells and vascular smooth muscle cells.
  • It contains a characteristic EGF-like domain with conserved disulfide bonds essential for its three-dimensional structure and receptor binding.
  • Subcellularly, BTC is localized to the cell membrane as a precursor and secreted into the extracellular space after processing.
  • BTC is predominantly expressed in pancreas and small intestine, but it is also synthesized in various other tissues and tumor cells, suggesting roles in both normal physiology and cancer.
  • Additional features include N-linked glycosylation and a single transmembrane helix, classifying it as a type I membrane protein.
  • The reference proteome entry highlights its identification by mass spectrometry-based proteomics, confirming endogenous expression.

Experimental Guidance and Technical Tips

  • The immunogen used to raise this antibody corresponds to the extracellular domain of human BTC (amino acids 32–178), which excludes the signal peptide (aa 1–31) and includes the putative mature growth factor region. Therefore, the antibody is expected to recognize both the full-length precursor and the cleaved mature protein.
  • For Western blot (WB), a band around 20 kDa is detected under reducing conditions, consistent with the predicted molecular weight of the mature BTC monomer. Consider using reducing and non-reducing conditions to observe disulfide-linked forms.
  • BTC is a secreted protein; thus, in cell lysates, it may be enriched in membrane fractions or concentrated in conditioned media. For tissue samples, pancreas and small intestine are recommended as positive controls.
  • In ELISA applications, this polyclonal antibody can serve as a capture or detection reagent, but optimal working concentrations and pairings should be validated in the relevant sample matrix.
