Biological Background: Barttin Function and Localization
- Barttin is a regulatory subunit of the anion-selective chloride channels CLCNKA and CLCNKB. These heteromeric channels are essential for basolateral chloride conductance along the nephron, enabling urine concentration and systemic acid-base homeostasis, and in the stria vascularis of the inner ear to establish the endocochlear potential necessary for normal hearing. Related references: PMID:11734858 PMID:12111250 PMID:12574213
- Barttin acts as a chaperone that allosterically regulates proper sorting of CLCNKA:BSND and CLCNKB:BSND channels to the basolateral plasma membrane domain and mediates the constitutive opening of channel common gates. Related references: PMID:11734858 PMID:12111250 PMID:12574213
- Encoded by the BSND gene, mutations in barttin are associated with Bartter syndrome type IV, characterized by renal salt wasting and sensorineural deafness.
- Barttin localizes specifically to the basolateral cell membrane in expressing tissues.
- It is predominantly expressed in the kidney, within specific nephron segments, and in the stria vascularis of the inner ear.
- Post-translational modifications include palmitoylation and phosphorylation, which may regulate its function and membrane association.
- The protein has a molecular weight of approximately 35 kDa.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 54-320 of human BSND, covering the majority of the protein. As a polyclonal antibody, it likely recognizes multiple epitopes; validate specificity in your sample system.
- For Western blot, a band around 35 kDa is expected. Use appropriate loading controls and secondary antibodies, and verify the band size with molecular weight markers.
- In ELISA applications, consider potential higher background common with polyclonal antibodies; optimize blocking and washing steps to improve signal-to-noise ratio.
- Reported cross-reactivity includes human, mouse, and rat; if working with other species or specific tissue types, confirm reactivity in your experimental context.
