Biological Background: BRRF2 Function and Localization

• BRRF2 is a tegument protein of Epstein-Barr virus (strain B95-8), also referred to as Human herpesvirus 4 (HHV-4). It is a structural component of the viral particle.
• The primary function of BRRF2 is to inhibit the dsDNA-triggered induction of host antiviral genes, a key immune evasion mechanism. It interacts with and inhibits host cyclic GMP-AMP synthase (cGAS), thereby limiting cGAS-DNA phase separation and downstream signaling.
• Subcellularly, BRRF2 is found in the virion tegument, localizes to host early endosomes, and is also secreted from infected cells.
• Post-translational modification: BRRF2 is a phosphoprotein, suggesting regulation by phosphorylation.
