Biological Background: Peregrin (BRPF1) Function and Localization
- BRPF1 (Bromodomain and PHD finger-containing protein 1), also known as Peregrin or Br140, is a scaffold subunit of histone acetyltransferase (HAT) complexes such as MOZ/MORF and HBO1, which acetylate histone H3 (PubMed:16387653, PubMed:24065767, PubMed:27939640). Related references: PMID:16387653 PMID:24065767 PMID:27939640
- Within the HBO1 complex, BRPF1 directs KAT7/HBO1 specificity toward histone H3 lysine-14 acetylation (H3K14ac) (PubMed:24065767). Related references: PMID:24065767
- Some BRPF1-containing HAT complexes also mediate histone H3 lysine-23 acetylation (H3K23ac) (PubMed:27939640). Related references: PMID:27939640
- BRPF1 positively regulates transcription of RUNX1 and RUNX2 (PubMed:18794358). Related references: PMID:18794358
- The protein contains multiple conserved domains, including a bromodomain, PHD finger, and zinc finger motifs, which mediate DNA binding and chromatin targeting.
- BRPF1 localizes predominantly to the nucleus and chromosomes, with some cytoplasmic presence.
- High expression is observed in the testis, suggesting tissue-specific functions.
- Post-translational modifications include acetylation and phosphorylation; mutations are associated with intellectual disability.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (aa 1-200) of human BRPF1, which may recognize full-length and certain splice variants; confirm specificity with appropriate controls.
- For Western blot (WB), the predicted molecular weight is ~137 kDa; use a suitable gel percentage and transfer conditions for high-molecular-weight proteins.
- In immunofluorescence/immunocytochemistry (IF/ICC), consider nuclear staining patterns consistent with chromatin association; validate with knockout or siRNA controls.
- The antibody reacts with human, mouse, and rat; for cross-species experiments, verify sequence homology in the immunogen region.
- ELISA can be used for antibody titer determination or antigen detection; optimize coating and blocking conditions.
