Biological Background: BRG1/SMARCA4 Function and Localization
- SMARCA4 (BRG1) is the SWI/SNF-related matrix-associated actin-dependent regulator of chromatin subfamily A member 4, also known as BAF190A, protein BRG-1, SNF2-beta, and transcription activator BRG1.
- Acts as an ATP-dependent chromatin remodeler, altering DNA-histone contacts to regulate transcription of select genes. Related references: PMID:15075294, PMID:29374058, PMID:30339381
- Core component of SWI/SNF chromatin remodeling complexes; in neural development, participates in the npBAF and nBAF complexes with subunit switching during differentiation.
- Functions as a corepressor of ZEB1 to regulate E-cadherin transcription and is required for TGF-beta-induced epithelial-mesenchymal transition (EMT). Related references: PMID:20418909
- Localizes to the nucleus, where it associates with chromatin and exerts its remodeling function.
- Colocalizes with ZEB1 in E-cadherin-negative cells of established lines, stroma of normal colon, and at the invasion front of colorectal carcinomas.

Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide corresponding to the N-terminal region (aa 30-130) of human SMARCA4; this region is relatively conserved and may detect multiple isoforms or related family members.
- Given the large molecular weight (185 kDa), use an SDS-PAGE gel with 6–8% polyacrylamide for optimal separation in western blotting; a low voltage transfer for extended time is recommended.
- For immunofluorescence/ICC, consider nuclear permeabilization and a nuclear counterstain to confirm localization; enrichment of nuclear fractions can enhance signal.
- Validate antibody specificity in the relevant cell or tissue system, especially if cross-reactivity with other species is expected, as official cross-reactivity data is limited to human.
- ELISA performance may depend on the antigen conformation; a competitive or sandwich format using the immunogen peptide as standard can be considered.
