Biological Background: BRAF Function and Localization
- BRAF (proto-oncogene B-Raf) is a serine/threonine-protein kinase that transduces mitogenic signals from the cell membrane to the nucleus.

- The immunogen peptide spans amino acids 507–606, covering the V600 codon; therefore, this antibody is designed to detect the BRAF V600E mutant.
- For western blotting, the expected molecular weight is ~84 kDa. Validate using human cell lines known to express the V600E mutation, alongside wild-type controls.
- For immunofluorescence and immunocytochemistry, optimize fixation and permeabilization conditions according to the cellular context.
- When performing ELISA, consider coating with the immunizing peptide or recombinant mutant BRAF protein to assess antibody specificity.
