Biological Background: BRAF Function and Localization

  • BRAF (proto-oncogene B-Raf) is a serine/threonine-protein kinase that transduces mitogenic signals from the cell membrane to the nucleus.
  • It phosphorylates MAP2K1, thereby activating the MAP kinase signal transduction pathway. Related references: PMID:21441910 / PMID:29433126
  • It phosphorylates PFKFB2, contributing to metabolic regulation. Related references: PMID:36402789
  • BRAF may play a role in the postsynaptic responses of hippocampal neurons. Related references: PMID:1508179
  • It is located in the nucleus, cytoplasm, and cell membrane.
  • BRAF is predominantly expressed in brain and testis.
  • The protein undergoes extensive regulatory modifications, including phosphorylation, acetylation, methylation, and ubiquitination, and contains a zinc-finger domain. The V600E mutation leads to constitutive kinase activity and is a major oncogenic driver.

Experimental Guidance and Technical Tips

  • The immunogen peptide spans amino acids 507–606, covering the V600 codon; therefore, this antibody is designed to detect the BRAF V600E mutant.
  • For western blotting, the expected molecular weight is ~84 kDa. Validate using human cell lines known to express the V600E mutation, alongside wild-type controls.
  • For immunofluorescence and immunocytochemistry, optimize fixation and permeabilization conditions according to the cellular context.
  • When performing ELISA, consider coating with the immunizing peptide or recombinant mutant BRAF protein to assess antibody specificity.
  • Cross-reactivity is reported for human; reactivity with other species should be tested individually.
