Biological Background: BNP Function and Localization

  • BNP (Natriuretic peptides B, also known as brain natriuretic factor prohormone or Iso-ANP) is a cardiac hormone that mediates cardio-renal homeostasis by binding NPR1, stimulating cGMP production, and regulating fluid-electrolyte balance through natriuresis, diuresis, vasorelaxation, and inhibition of renin and aldosterone secretion. It also binds the clearance receptor NPR3. Related references: PMID:1672777, PMID:17372040, PMID:1914098, PMID:17349887, PMID:9458824, PMID:16870210

  • Processed forms such as brain natriuretic peptide 32 exhibit low potency in cGMP production but may still contribute to cardio-renal homeostasis. Related references: PMID:17372040

  • BNP is secreted and predominantly detected in cardiac atria and kidney distal tubular cells, with additional expression in bone growth plate chondrocytes (late proliferative to hypertrophic). Related references: PMID:2136732, PMID:2138890, PMID:9794555

  • As a member of the natriuretic peptide family, BNP features a disulfide bond, glycosylation, and proteoglycan modifications, and acts as a vasodilator.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to a sequence within the propeptide region (aa 35-134). This antibody may detect both proBNP and processed BNP peptides in WB. Due to the low molecular weight of mature BNP (~3.5 kDa), careful gel conditions and transfer settings are recommended.
  • For ELISA applications, consider pairing with capture antibodies recognizing distinct epitopes for sandwich detection.
  • Validate specificity in relevant human, mouse, or rat cardiac tissue or plasma samples, as cross-reactivity has been reported.
