Biological Background: BNIP1 Function and Localization
- BNIP1 (also known as SEC20, NIP1, and TRG-8) is a vesicle transport protein that functions as part of a SNARE complex involved in endoplasmic reticulum (ER) membrane fusion and maintenance of ER organization. Related references: PMID:15272311
- BNIP1 plays a role in apoptosis, including being required for endoplasmic reticulum stress-induced apoptosis. Related references: PMID:15272311 PMID:23896122 PMID:7954800
- As a substrate of RNF185 interacting with SQSTM1, BNIP1 might be involved in mitochondrial autophagy.
- BNIP1 is localized to the endoplasmic reticulum membrane and the mitochondrial membrane.
- Alternative splicing produces multiple isoforms with distinct tissue expression patterns: isoform 1 is highly expressed in heart, brain, liver, skeletal muscle, and pancreas; isoform 3 in placenta, lung, and kidney; isoform 4 in testis and small intestine.
- The protein contains a coiled-coil domain, a transmembrane helix, and is subject to ubiquitin conjugation. It is also involved in ER-Golgi transport and host-virus interactions.
Experimental Guidance and Technical Tips
- As a polyclonal antibody, this product may detect multiple BNIP1 isoforms; consider validating reactivity in your specific sample system.
- For Western blot, a band of approximately 26 kDa is expected; optimize blocking and detection conditions for polyclonal antibodies.
- The immunogen comprises the N-terminal region (amino acids 1–200), making the antibody suitable for recognizing full-length BNIP1.
- Due to cross-reactivity with mouse and rat, this antibody can be used in those species with appropriate validation.
- For ELISA, consider pairing with a suitable detection antibody and optimizing coating conditions.
