Biological Background: BMPR1A Function and Localization
- BMPR1A encodes the bone morphogenetic protein receptor type-1A, a single-pass transmembrane serine/threonine kinase also designated ALK-3, SKR5, and CD292.
- Upon ligand binding, BMPR1A assembles into a heterotetrameric complex with type II receptors, which phosphorylate and activate the type I receptor; activated BMPR1A then phosphorylates SMAD transcriptional regulators to modulate target gene expression.
- It serves as the primary receptor for BMP2, BMP4, GDF5, and GDF6 ligands, and positively regulates chondrocyte differentiation through GDF5 signaling and promotes adipogenesis in response to GDF6.
- BMPR1A may also upregulate HAMP expression, likely through its interaction with BMP2 (by similarity).
- The receptor localizes to the cell membrane and cell surface, consistent with its role as an extracellular ligand-sensing kinase.
- BMPR1A contains an ATP-binding kinase domain, disulfide bonds, glycosylation sites, and ubiquitination motifs, and is classified under several key functional keywords including serine/threonine-protein kinase, receptor, and transmembrane signaling.
- It is highly expressed in skeletal muscle, indicating a prominent role in muscle homeostasis or development.
Experimental Guidance and Technical Tips
- When using this polyclonal antibody in Western blot, a band near 60 kDa is expected; consider optimizing lysis and denaturing conditions to ensure full epitope exposure.
- For ELISA applications, pairing with a suitable detection antibody may enhance sensitivity; validate pairings in the relevant sample matrix.
- Cross-reactivity with mouse and rat BMPR1A has been noted; however, sequence alignment and experimental validation in the species of interest is recommended to confirm specificity.
