Biological Background: BMP6 Function and Localization
- Bone morphogenetic protein 6 (BMP6), also designated VG-1-related protein (VGR-1), is a member of the TGF-beta superfamily and plays essential roles in cartilage and bone formation. Related references: PMID:31019025
- BMP6 is a key regulator of hepcidin (HAMP) expression and iron metabolism, acting as a ligand for hemojuvelin/HJV and signaling through BMPR1A/ALK3 to promote HAMP transcription. Related references: PMID:26582087 PMID:30097509 PMID:31800957
- It initiates canonical BMP signaling by associating with type I receptor ACVR1 and type II receptor ACVR2B, leading to phosphorylation of SMAD1/5/8 and transcriptional regulation. Related references: PMID:18070108
- BMP6 can also signal through non-canonical TAZ-Hippo pathway to modulate VEGF signaling by regulating VEGFR2 expression. Related references: PMID:33021694
- Subcellular localization: secreted glycoprotein that undergoes cleavage on a pair of basic residues and contains disulfide bonds.
- Keywords: chondrogenesis, osteogenesis, developmental protein, cytokine, growth factor.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 350–450 of human BMP6 (NP_001709.1), a region that may be conserved across human, mouse, and rat, consistent with the antibody's reported cross reactivity. Consider sequence alignment to verify epitope conservation.
- For Western blot (WB), a reducing and denaturing sample preparation is recommended to detect the expected ~57 kDa monomer. Non-reduced samples may show higher molecular weight forms due to disulfide bonds.
- In immunofluorescence/immunocytochemistry (IF/ICC), validate antibody specificity using appropriate controls, such as knockdown or overexpression cell lines, to confirm localization.
- For ELISA, the antibody can be explored as a capture or detection reagent in sandwich assays. Pairing with another BMP6-specific antibody may be necessary for optimal performance.
