Biological Background: BMP5 Function and Localization
- BMP5 (bone morphogenetic protein 5) is a secreted glycoprotein and member of the TGF-beta superfamily, encoded by the BMP5 gene. Alternative splicing produces multiple isoforms, and the protein undergoes proteolytic cleavage.
- BMP5 plays essential roles in developmental processes, including cartilage and bone formation and neurogenesis. Related references: PMID:11580864, PMID:29321139
- It initiates canonical BMP signaling by binding to type I receptor BMPR1A and type II receptor BMPR2, leading to phosphorylation of SMAD1/5/8 transcription factors. Related references: PMID:11580864, PMID:29321139
- BMP5 can also signal through the non-canonical MAPK p38 pathway to promote chondrogenic differentiation. Related references: PMID:20402566
- It promotes the expression of HAMP (hepcidin), and this activity is repressed by interaction with ERFE (erythroferrone). Related references: PMID:30097509
- Subcellular localization: BMP5 is secreted and acts as an extracellular signaling molecule.
- Tissue specificity: Expressed in the lung and liver.
- Keywords: Chondrogenesis, osteogenesis, neurogenesis, cytokine, developmental protein, differentiation, disulfide bond, glycoprotein.
Experimental Guidance and Technical Tips
- The immunogen used to raise this antibody corresponds to amino acids 317–454 of human BMP5, a region in the C-terminal portion of the protein. This design may confer target specificity, but cross-reactivity with other BMP family members should be evaluated in relevant assay systems.
- For western blot (WB), a band around 52 kDa is expected; however, actual migration may vary due to post-translational modifications. Use appropriate positive controls (e.g., BMP5-expressing cell lysates or recombinant protein) and validate in the relevant species (human, mouse, rat).
- In immunohistochemistry on paraffin-embedded sections (IHC-P), consider optimizing antigen retrieval conditions and antibody dilution. Tissue sections from lung or liver (where BMP5 is expressed) may serve as positive control.
- For ELISA applications, confirm reactivity against the immunogen or full-length BMP5 protein and establish standard curves with known concentrations.
