Biological Background: BMP4 Function and Localization
- BMP4 (bone morphogenetic protein 4), also designated BMP-2B, is a secreted cytokine of the transforming growth factor-beta (TGF-beta) superfamily that orchestrates embryonic development, including neurogenesis, vascular formation, and bone morphogenesis.
Related references: PMID:31363885 - The protein initiates canonical signaling by assembling a receptor complex with BMPR1A and BMPR2, which phosphorylates SMAD1/5/8 transcription factors to regulate target gene expression.
Related references: PMID:25868050 PMID:8006002 PMID:29212066 - BMP4 also activates non-SMAD pathways, such as ERK/MAP kinase, PI3K/Akt, and SRC cascades; SRC-mediated VEGFR2 phosphorylation promotes an angiogenic response.
Related references: PMID:31363885 - After synthesis, BMP4 is proteolytically cleaved and secreted into the extracellular matrix where it functions as a mature growth factor; disulfide bonds and glycosylation are critical for its structural stability and bioactivity.
- The protein is predominantly expressed in the lung, with lower levels in the kidney, and is detected in normal and cancerous prostate tissues.
- BMP4 drives odontogenesis by inducing MSX1-mediated transcription and Wnt signaling in dental mesenchymal cells, essential for tooth bud progression, while later being repressed to allow terminal differentiation.
- Beyond development, BMP4 cooperates with PTHLH to stimulate mammary ductal outgrowth and suppresses hair follicle induction.
Experimental Guidance and Technical Tips
- The immunogen was designed from a C-terminal peptide (residues 309–408) that is highly conserved in human BMP4, providing targeted antibody specificity.
- When performing Western blot assays, expect a protein band near 47 kDa under standard reducing conditions.
- For immunohistochemistry on paraffin-embedded tissues, consider heat-induced epitope retrieval to expose antigenic sites effectively.
- Since mature BMP4 is secreted, immunofluorescence detection of intracellular antigen may require cell permeabilization to visualize precursor pools.
- The antibody is specific for human BMP4; cross-reactivity with other species remains to be tested and should be validated by the end user.
