Biological Background: BMP4 Function and Localization

  • BMP4 (Bone Morphogenetic Protein 4), also known as BMP-2B, is a secreted growth factor belonging to the TGF-beta superfamily. It plays critical roles in developmental processes including neurogenesis, angiogenesis, and osteogenesis. Related references: PMID:31363885
  • The protein initiates canonical BMP signaling by binding to type I receptor BMPR1A and type II receptor BMPR2, leading to phosphorylation and activation of SMAD1/5/8 transcription factors. Related references: PMID:25868050, PMID:8006002, PMID:29212066
  • It can also activate non-canonical pathways such as ERK/MAP kinase, PI3K/Akt, or SRC cascades to mediate angiogenic responses. Related references: PMID:31363885
  • BMP4 positively regulates odontogenic development via MSX1-mediated Wnt signaling and plays a role in mammary gland development and hair follicle inhibition.
  • Subcellular localization: BMP4 is secreted into the extracellular space and associates with the extracellular matrix.
  • Tissue specificity: It is expressed predominantly in the lung and at lower levels in the kidney; also found in normal and neoplastic prostate tissues and prostate cancer cell lines.

Experimental Guidance and Technical Tips

  • The immunogen comprises a synthetic peptide spanning residues 250–350 of human BMP4. This region is likely surface-exposed in the mature dimeric protein, enabling detection of full-length endogenous BMP4 under appropriate conditions.
  • For Western blotting, BMP4 runs as a ~47 kDa monomer under reducing conditions; under non-reducing conditions, a disulfide-linked homodimer band at ~94 kDa may be observed. Include both conditions to confirm specificity.
  • Validate antibody performance in lysates from BMP4-expressing tissues such as lung or kidney, or in cell lines derived from prostate cancer.
  • For ELISA, consider pairing this antibody with a BMP4 capture or detection antibody in a sandwich format, ensuring the pair recognizes distinct epitopes.
