Biological Background: BMP4 Function and Localization
- BMP4 (Bone Morphogenetic Protein 4), also known as BMP-2B, is a secreted growth factor belonging to the TGF-beta superfamily. It plays critical roles in developmental processes including neurogenesis, angiogenesis, and osteogenesis. Related references: PMID:31363885

- The immunogen comprises a synthetic peptide spanning residues 250–350 of human BMP4. This region is likely surface-exposed in the mature dimeric protein, enabling detection of full-length endogenous BMP4 under appropriate conditions.
- For Western blotting, BMP4 runs as a ~47 kDa monomer under reducing conditions; under non-reducing conditions, a disulfide-linked homodimer band at ~94 kDa may be observed. Include both conditions to confirm specificity.
- Validate antibody performance in lysates from BMP4-expressing tissues such as lung or kidney, or in cell lines derived from prostate cancer.
- For ELISA, consider pairing this antibody with a BMP4 capture or detection antibody in a sandwich format, ensuring the pair recognizes distinct epitopes.
