Biological Background: BMP4 Function and Localization

  • BMP4 (bone morphogenetic protein 4), also known as BMP-2B, is a secreted growth factor belonging to the TGF-beta superfamily that plays critical roles in embryonic development, including neurogenesis, vascular development, angiogenesis, and osteogenesis. Related references: PMID:31363885

  • This polyclonal antibody was generated using a synthetic peptide corresponding to amino acids 250-350 of human BMP4 (NP_001193.2). For optimal results, consider validating antibody specificity in your experimental system using positive and negative controls.
  • In Western blotting (WB), the expected molecular weight of BMP4 is approximately 47 kDa under reducing conditions. A band near this size may be observed; however, additional bands corresponding to precursor or cleaved forms might appear due to proteolytic processing.
  • For immunohistochemistry (IHC-P), antigen retrieval methods (e.g., heat-induced epitope retrieval) are recommended to expose the epitope. Optimize antibody dilution and incubation conditions for your specific tissue type.
  • In immunofluorescence (IF/ICC), include appropriate controls to distinguish specific staining from background. Permeabilization may be necessary for intracellular staining.
  • For ELISA applications, the antibody can be used as a capture or detection reagent. Pair it with a validated matched secondary antibody for detection.
