Biological Background: BMP2 Function and Localization
- BMP2 (Bone morphogenetic protein 2), also known as BMP-2A, is a member of the TGF-beta superfamily that plays essential roles in developmental processes, including cardiogenesis, neurogenesis, and osteogenesis. Related references: PMID:18436533, PMID:24362451, PMID:31019025
- It induces cartilage and bone formation by initiating the canonical BMP signaling cascade through association with type I receptor BMPR1A and type II receptor BMPR2. Related references: PMID:3201241, PMID:15064755, PMID:17295905
- Upon receptor binding, BMPR2 phosphorylates and activates BMPR1A, which then phosphorylates SMAD1/5/8 to regulate transcription of target genes. Related references: PMID:7791754
- BMP2 can also signal through non-canonical pathways such as ERK/MAP kinase signaling to regulate osteoblast differentiation. Related references: PMID:16771708, PMID:20851880
- It stimulates myoblast differentiation into osteoblasts via the EIF2AK3-EIF2A-ATF4 pathway, increasing ATF4 expression. Related references: PMID:24362451
- The protein is secreted and exhibits widespread tissue expression, with particular abundance in lung, spleen, and colon; it is also present in heart, brain, placenta, liver, kidney, and pancreas.
- BMP2 acts as a positive regulator of odontoblast differentiation during tooth germ formation and promotes HAMP expression through BMPR1A/ALK3. Related references: PMID:31800957
Experimental Guidance and Technical Tips
- The immunogen corresponds to a recombinant fusion protein containing amino acids 283-396 of human BMP2; consider potential cross-reactivity with other BMP family members sharing homologous regions.
- In Western blot, the full-length BMP2 appears as a ~45kDa band, while the mature active form is approximately 18kDa; validate detection in your specific samples.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), antigen retrieval and optimization of antibody dilution and detection conditions may be necessary due to the secreted nature of BMP2.
- ELISA applications can utilize this antibody as capture or detection; confirm optimal working concentrations and compatibility with your assay format.
- When working with mouse or rat samples, validate reactivity given high sequence similarity; positive controls are recommended.
