Biological Background: BMP1 Function and Localization
-
Bone morphogenetic protein 1 (BMP1) is a metalloprotease that processes various precursor proteins into mature functional enzymes or structural proteins, playing a critical role in regulating the formation of the extracellular matrix (ECM). This activity is essential for cartilage and bone formation, muscle growth, wound healing, and tissue repair. Related references: PMID:33206546 PMID:32636307 PMID:33169406
-
BMP1 cleaves the C-terminal propeptides from procollagens I, II, and III and proteolytically activates lysyl oxidase (LOX), which is required for covalent cross-linking in collagen and elastic fibers. Related references: PMID:31152061 PMID:33206546
-
The enzyme also processes matricellular thrombospondin-1 (THBS1), leading to disruption of cell adhesion and activation of TGF-beta signaling. Related references: PMID:32636307
-
BMP1 serves as a coactivator of BMP7, contributing to bone repair processes.
-
Alternative splicing generates isoforms, including the longer mammalian tolloid protein (mTld), which shares functional domains.
-
Subcellular localization includes the Golgi apparatus (trans-Golgi network) and secretion into the extracellular space and extracellular matrix.
-
Ubiquitous tissue expression highlights its broad role in ECM maintenance and developmental processes.
Experimental Guidance and Technical Tips
- The immunogen corresponds to residues 725–986 of human BMP1, a region likely conserved across isoforms; note that polyclonal antibodies may recognize multiple isoforms.
- Recommended applications are IF/ICC and ELISA; for IF/ICC, consider optimizing fixation and permeabilization conditions due to the secreted and Golgi-localized nature of BMP1.
- For ELISA, validate antibody specificity using recombinant BMP1 protein or known positive controls, as cross-reactivity with closely related metalloproteases is possible with polyclonal antibodies.
- Protein molecular weight is 111 kDa; expect bands near this size in western blotting if additional validation is performed, but note that the antibody is not validated for WB.
