Biological Background: BMAL1 Function and Localization
- BMAL1 (ARNTL) is a basic helix-loop-helix PAS family transcription factor that forms a core component of the circadian clock. It heterodimerizes with CLOCK or NPAS2 to drive the expression of clock genes and clock-controlled genes through E-box elements, regulating daily physiological rhythms.
- The CLOCK-BMAL1 heterodimer recognizes canonical (5′-CACGTG-3′) and non-canonical E-box motifs, with a preferred binding motif of 5′-CACGTGA-3′. BMAL1 binds the GTGA half-site while CLOCK binds CAC (PubMed:23229515). Related references: PMID:23229515
- BMAL1 plays a critical role in rhythmic chromatin opening, and the BMAL1-CLOCK complex regulates the circadian expression of numerous genes involved in metabolism, including SERPINE1, NAMPT, DBP, and PPARGC1A, linking the clock to metabolic homeostasis.

- The immunogen corresponds to amino acids 380–510, a region outside the bHLH and PAS domains; this choice may reduce cross-reactivity with other bHLH-PAS proteins and enhance detection of full-length BMAL1.
- For Western blotting, an expected band near 69 kDa should be observed in human, mouse, and rat cell extracts (e.g., HeLa, mouse brain, rat liver). Use appropriate loading controls and consider nuclear extraction to enrich the signal.
- For ELISA applications, optimize coating concentration of the recombinant immunogen or native protein, and validate specificity using competition assays or knockout models.
- Cross-reactivity with mouse and rat is anticipated; however, sequence alignment of the immunogen region across species is recommended to confirm epitope conservation.
