Biological Background: BLOC1S2 Function and Localization
- BLOC1S2, also known as centrosome-associated protein (CEAP), BLOS2, or BORCS2, is a component of the biogenesis of lysosome-related organelles complex 1 (BLOC-1) that is essential for normal biogenesis of lysosome-related organelles (LROs) such as platelet dense granules and melanosomes. - In neurons, BLOC-1 cooperates with the AP-3 complex to target membrane protein cargos into vesicles assembled at cell bodies for delivery into neurites and nerve terminals, and is also proposed to be involved in neurite extension.
- BLOC1S2 is also a subunit of the BORC complex, which associates with the cytosolic face of lysosomes and recruits ARL8B to couple lysosomes to microtubule plus-end-directed kinesin motor, thereby regulating lysosome movement and peripheral localization. Related references: PMID:25898167
