Biological Background: BIRC7 Function and Localization
- BIRC7 is an apoptotic regulator with both pro- and anti-apoptotic activities, playing crucial roles in apoptosis, cell proliferation, and cell cycle control. Related references: PMID:11024045 PMID:11084335 PMID:11162435
- Its anti-apoptotic activity is mediated by inhibition of CASP3, CASP7, and CASP9, as well as by its E3 ubiquitin-protein ligase activity, which targets DIABLO/SMAC for degradation, promoting cell survival. Related references: PMID:11024045 PMID:16729033
- It protects against apoptosis induced by TNF, adriamycin, etoposide, or staurosporine; suppression involves activation of MAPK8/JNK1 and possibly MAPK9/JNK2 via TAB1 and MAP3K7/TAK1. Related references: PMID:11084335 PMID:11162435 PMID:11865055
- Blocks staurosporine- and etoposide-induced apoptosis, and modulates natural killer (NK) cell-mediated killing. Related references: PMID:11322947 PMID:18034418 PMID:11162435
- Subcellularly, BIRC7 localizes to the nucleus, cytoplasm, and Golgi apparatus.
- Isoform 1 and isoform 2 are expressed at very low levels in most adult tissues but are detected in heart, placenta, lung, lymph node, spleen, ovary, and several carcinoma cell lines.
- Contains a BIR domain and a RING-type zinc finger, characteristic of both apoptosis inhibitors and E3 ubiquitin ligases.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant protein spanning amino acids 1–265 of human BIRC7. Consider using whole-cell lysates from BIRC7-expressing cell lines (e.g., HeLa, MCF7) as positive controls for WB.
- For WB, optimize primary antibody dilution and validate specificity via siRNA-mediated knockdown or CRISPR knockout of BIRC7.
- For ELISA applications, pair with an appropriate capture/detection antibody and test under various coating conditions to achieve optimal signal-to-noise ratios.
