Biological Background: BIRC7 Function and Localization

  • BIRC7, also known as Livin, KIAP, ML-IAP, or RNF50, is a member of the inhibitor of apoptosis (IAP) family that contains a baculoviral IAP repeat (BIR) domain and a RING finger domain.
  • BIRC7 is a dual-function apoptotic regulator, exerting both pro- and anti-apoptotic activities; its anti-apoptotic function involves inhibition of CASP3, CASP7, and CASP9, and E3 ubiquitin-protein ligase-mediated degradation of SMAC/DIABLO to relieve XIAP inhibition. Related references: PMID:11024045, PMID:16729033
  • It protects cells from apoptosis induced by TNF, adriamycin, etoposide, or staurosporine, with suppression mediated by MAPK8/JNK1 and MAPK9/JNK2 activation via TAB1 and MAP3K7/TAK1. Related references: PMID:11084335, PMID:11162435, PMID:11865055
  • The protein additionally blocks staurosporine- and etoposide-induced apoptosis and modulates natural killer cell-mediated cytotoxicity. Related references: PMID:11322947, PMID:11162435, PMID:18034418
  • It localizes to the nucleus, cytoplasm, and Golgi apparatus.
  • Isoforms 1 and 2 show low or undetectable expression in most adult tissues but are detected in placenta, lung, lymph node, spleen, ovary, and various carcinoma cell lines; isoform 2 is additionally found in fetal kidney, heart, and spleen.
  • BIRC7 is classified as a protease inhibitor and thiol protease inhibitor; it contains zinc-finger motifs and participates in ubiquitin conjugation pathways.

Experimental Guidance and Technical Tips

  • The immunogen (aa 1-265) encompasses the BIR domain, likely generating antibodies that recognize both BIRC7 isoforms; verify isoform specificity if required for your experimental design.
  • For Western blotting, expect a band at ~33 kDa; consider using a low-percentage gel to resolve close molecular weight species and validate with appropriate controls (e.g., siRNA knockdown or overexpression lysates).
  • This polyclonal antibody is qualified for WB, IHC-P, IF/ICC, and ELISA. For immunohistochemistry, optimize antigen retrieval and antibody dilution using positive control tissues such as placenta or carcinoma samples known to express BIRC7.
  • Cross-reactivity with mouse and rat enables use in rodent models; confirm reactivity in your specific sample context.
  • As BIRC7 expression is generally low in normal tissues, employ cancer cell lines (e.g., melanoma, lung carcinoma) with upregulated Livin as positive controls.
