Biological Background: BiP/GRP78 Function and Localization
- Endoplasmic reticulum chaperone that plays a key role in protein folding and quality control in the ER lumen. Related references: PMID:2294010, PMID:23769672, PMID:23990668
- Involved in the degradation of misfolded proteins through interaction with DNAJC10/ERdj5.
- Acts as a key repressor of the unfolded protein response (UPR) by binding and inactivating EIF2AK3/PERK and ERN1/IRE1 under non-stress conditions. Related references: PMID:11907036, PMID:1550958, PMID:19538957
- May function as an allosteric modulator for SEC61 translocon, regulating post-translational translocation of small presecretory proteins.
- Subcellularly localized predominantly to the ER lumen, but also detected in melanosomes, cytoplasm, and cell surface under specific conditions.
- Serves as a cell surface receptor for Dengue virus, Zika virus, and Japanese encephalitis virus, facilitating viral entry. Related references: PMID:15098107, PMID:28053106, PMID:33432092
- Acts as a receptor for CotH proteins from mucorales fungi, mediating epithelial invasion and contributing to mucormycosis pathogenesis. Related references: PMID:20484814, PMID:24355926, PMID:32487760
- Post-translationally modified by acetylation, methylation, nitration, phosphorylation, and ubiquitination; possesses an ATP-binding domain characteristic of HSP70 family.
Experimental Guidance and Technical Tips
- The immunogen, a synthetic peptide mapping to amino acids 550–654 within the C-terminal region of human BiP/GRP78, is expected to generate antibodies recognizing native and denatured protein, suitable for multiple applications.
- For Western blot analysis, a band corresponding to the predicted molecular weight of approximately 72 kDa should be observed; additional bands may appear due to post-translational modifications or degradation.
- For immunofluorescence and immunohistochemistry, the predominant ER staining pattern is anticipated; however, under stress conditions or in certain cell types, cell surface localization may also be detectable.
- Cross-reactivity with mouse and rat BiP/GRP78 is reported; validate the antibody in the relevant cell or tissue lysate before use.
- In ELISA applications, this polyclonal antibody can serve as a capture or detection reagent, but optimal working dilutions must be empirically determined.
